Possible mechanisms of Siddha Anti-Cancer Medicines
Thiripala Choornam has demonstrated significant cytotoxic and antiproliferative activity against various cancer cell lines, including cervical adenocarcinoma, endometrial adenocarcinoma, breast adenocarcinoma, oral squamous cell carcinoma, lung carcinoma, hepatocellular carcinoma, prostate cancer, ovarian cancer, and pancreatic cancer. Beyond its cytotoxic effects, mechanistic studies have shown that it can induce apoptosis, regulate cell-cycle arrest, modulate mitochondrial pathways, alter Bax/Bcl-2 expression ratios, activate caspases, and suppress invasive and metastatic behaviour of cancer cells.
2. Rasam related formulations may induce intracellular oxidative stress, leading to mitochondrial dysfunction and activation of the intrinsic apoptotic pathway. This process involves the release and enhancement of cytochrome-c from mitochondria, which subsequently activates the caspase pathway. Caspases, the intracellular enzymes responsible for apoptosis, promote DNA damage and ultimately lead cancer cells to undergo programmed cell death.
3. Thamiram based formulations may elevate intracellular copper levels, thereby disrupting cancer cell homeostasis and promoting excessive generation of reactive oxygen species (ROS). The resulting sustained redox imbalance can induce mitochondrial dysfunction, lipid peroxidation, and DNA damage, ultimately inhibiting tumour cell proliferation and promoting cancer cell death.
